誘導(dǎo)過程及結(jié)果評估

給實(shí)驗(yàn)組和對照組小鼠分別喂食維生素D缺乏飼料及充足飼料6周,然后改為高脂肪維生素D缺乏飼料及充足飼料8-10周。飲食誘導(dǎo)的維生素D缺乏小鼠收縮壓和舒張壓升高,血漿腎素升高,尿鈉排泄減少。隨后給維生素D缺乏小鼠重新喂食維生素D充足飼料6周,高血壓得到逆轉(zhuǎn),腎素得到抑制。

在高脂飲食過程中,與維生素D充足的小鼠相比,維生素D缺乏小鼠的主動脈弓動脈粥樣硬化加快2倍,胸腹主動脈動脈粥樣硬化加快2-8倍。在主動脈根部,高脂飼料喂養(yǎng)的維生素D缺乏小鼠的巨噬細(xì)胞浸潤增加,脂肪積聚和內(nèi)質(zhì)網(wǎng)(ER)應(yīng)激激活增加,但動脈粥樣硬化斑塊內(nèi)M1巨噬細(xì)胞表型的發(fā)生率較低。同樣,維生素D缺乏小鼠腹腔巨噬細(xì)胞表現(xiàn)出M2為主的表型,泡沫細(xì)胞形成和內(nèi)質(zhì)網(wǎng)應(yīng)激增加。高脂飼料喂養(yǎng)期間用內(nèi)質(zhì)網(wǎng)應(yīng)激緩解劑PBA治療維生素D缺乏小鼠可抑制動脈粥樣硬化,減少腹腔巨噬細(xì)胞泡沫細(xì)胞的形成,下調(diào)內(nèi)質(zhì)網(wǎng)應(yīng)激蛋白而不改變血壓。

因此,我們認(rèn)為維生素D缺乏會激活腎素血管緊張素系統(tǒng)和巨噬細(xì)胞內(nèi)質(zhì)網(wǎng)應(yīng)激,從而促進(jìn)高血壓的發(fā)展和加速動脈粥樣硬化,維生素D替代療法是一種潛在的降壓和動脈粥樣硬化治療方法。
